Abstract
Pancreatic cancer is the 8th most common cause of cancer death for men and 9th for women worldwide with an incidence of approximately 10/100.000. Pancreatic ductal adenocarcinoma (PDAC) represents 85% of all cases of pancreatic cancer. Despite the constant efforts to develop novel diagnostic tools and treatment approaches, it is projected that PDAC would rise to the second most common cause of cancer death by 2030. Surgical resection, followed by adjuvant therapy, is the only radical therapeutic option. The prognosis of PDAC patients remains unfavorable for all stages with the 5-year survival rate being <5%. Aggressive tumor biology, as well as primary or secondary drug resistance, contribute significantly to this dismal prognosis. The main objective of this proposal is to generate the proof of principle for the use of sigma 2 receptors through their ligands as novel targets for the development of more potent and safer therapeutic modalities for the treatment of PDAC. Towards this aim a) we’ll study the largely unknown mechanism of action of these ligands in PDAC b) we will develop innovative nanostructures for drug delivery (DDN) such as linear pentablock hybrid quarterpolypeptides, using the Ring- Opening Polymerization (ROP) of N-carboxy anhydrides (ΝCAs) and nanoparticles (NPs) such as liposomes and liquid crystals for the targeted delivery of sigma ligands as monotherapy or in combination with standards of care for PDAC treatment (e.g. gemcitabine and paclitaxel) aiming to increase the efficacy and reduce any unwanted effects c) we will obtain the proof-of-principle by evaluating the activity DDN carrying the selected ligands in mouse models of cancer. In this last approach, we will use mainly patient-derived xenografts, either ectopic or orthotopic (PDOX) which are considered the most clinically relevant models. We need to emphasize that none of the above-mentioned scientific questions have been addressed worldwide, making this proposal unique in the field.
